trans-[Pt(OBz)2(O,C-10-BzODA)(1R,2R-DACH)] is an experimental platinum(IV) prodrug and analog of oxaliplatin designed for the treatment of pancreatic cancer. It features a (1R,2R)-diaminocyclohexane (DACH) carrier ligand, two axial benzoate ligands, and a 10-benzoyloxy-2-decenoate (10-BzODA) ligand. This complex is characterized by its ability to accumulate intracellularly without premature reduction in the extracellular environment. Its unique mechanism of action includes the induction of mitochondrial hyperpolarization and caspase-dependent apoptosis, distinguishing it from conventional platinum(II) drugs like cisplatin and oxaliplatin. Preclinical studies in pancreatic cancer cell models and 3D tumor models have shown it to possess superior antiproliferative efficacy compared to standard platinum therapies.
